Amorphical: Interview With CEO Eden Ben About The Biotechnology Company

Amorphical is a biotechnology company that develops nature-inspired therapeutics and mineral-based dietary supplements, focusing specifically on stabilizing and utilizing Amorphous Calcium Carbonate (ACC) to improve mineral absorption and treat major chronic, inflammatory, and oncological diseases. Could you tell me more about your background? Ben said:

I am Eden Ben. A biomedical engineer by training, I’ve grown with Amorphical from the ground up, starting in production operations and advancing through multiple roles before becoming CEO in 2025. This journey has given me a deep, hands-on understanding of every aspect of the company – from manufacturing and operations to R&D and business development.

This hands-on experience, together with my academic background, allows me to bring both the technical insight to evaluate our science and the practical experience to lead our people and operations. This unique perspective allows me to bridge strategy with execution and to guide Amorphical with both vision and pragmatism.

Formation Of The Company

How did the idea for the company come together? Ben shared:

Amorphical began in 2004 with a fascinating observation by one of its founders, Yossi Ben, a farmer who raised antelope and zebras and cultivated crayfish for aquarium displays. He was intrigued by the crayfish’s remarkable ability to regenerate their exoskeletons after shedding. These crayfish can regrow their shells in just three days, more than nine times faster than their lobster counterparts. After researching further, he discovered that crayfish deposit gastroliths, which are small, stone-like structures, in their stomachs when shedding. These gastroliths secrete an amorphous calcium carbonate that significantly accelerates shell growth.

Working with founding member Dr. Yigal Blum, they synthesized a proprietary mineral that mimics the unique structural profile of the crayfish’s amorphous calcium carbonate (ACC). They found that the ACC is absorbed more efficiently into the body than traditional crystalline calcium supplements, offering a unique treatment route.

Amorphical initially offered the ACC for bone degeneration, but soon observed consistent benefits extending far beyond that focus. This evidence revealed a profound insight: the mineral’s potential went far deeper, showing signs of disease-modifying effects.

Driven by this discovery, the company embarked on a pivotal mission: to synthesize the mineral in its rare nano-amorphous form for precise, therapeutic-level dosing.

Favorite Memory

What has been your favorite memory working for the company so far? Ben reflected:

One of my favorite memories was during my time in the lab, when I had an idea about how we could formulate our active ingredient to treat a specific type of cancer. After developing the concept, we tested it in a preclinical model, and the results ultimately led to a new patent.

What made that moment so meaningful wasn’t just the scientific insight, but the realization that I could connect different pieces of knowledge, translate them into a practical solution and then see that solution evolve into real business value. It was the point where science, creativity, and strategy came together — and that’s something I’ll never forget.

Core Products

What are the company’s core products and features? Ben explained:

We are developing a nano-amorphous mineral platform that offers superior bioavailability and a unique pH-responsive therapeutic mechanism. Remaining intact during circulation, these molecules dissolve in acidic microenvironments commonly found in diseased tissues. This release of calcium and carbonate ions modulates pathological sites, offering broad therapeutic applications across bone health, metabolic disease, inflammatory disease and oncology.

Amorphical currently has four therapeutics in clinical development:

  • Our lead program is in hypoparathyroidism which is being studied in an FDA Phase 2b trial. The nano-amorphous therapy aims to maintain stable calcium levels with minimal side effects.
  • We’re also studying an ACC treatment in a Phase 1/2 trial for osteoporotic fractures, which aims to accelerate bone healing and reduce pain.
  • We recently completed a Phase 1/2 in Crohn’s disease, showing that the therapy achieved sustained clinical remission at 24 weeks without the need for immunosuppression. We’re planning a larger Phase 2 trial to continue to study the impact in this disease.
  • Finally, we have an ongoing Phase 1/2 trial in Stage IV pancreatic cancer that buffers the tumor’s acidic microenvironment to enhance treatment response.

Challenges Faced

Have you faced any challenges in your sector of work recently? Ben acknowledged:

The most significant challenge we have overcome was stabilizing our nano-amorphous mineral technology for pharmaceutical use, which took over a decade of dedicated research. The natural form of this technology, found in the blue crayfish, is highly unstable outside its biological environment, so we needed to develop a method to maintain its amorphous, nanometric structure to ensure its superior bioavailability.

This involved extensive experimentation, focusing on different types and amounts of stabilizers that are readily available in the market. Crucially, we developed a very complex process that gave us the ability to stabilize it. This breakthrough allowed us to transition from the extremely costly and unsustainable method of extracting this mineral from crayfish to a highly efficient, chemically produced method in our lab and factory, significantly reducing production costs and enabling scalable manufacturing.

Evolution Of The Company’s Technology

How has the company’s technology evolved since launching? Ben noted:

*When we first synthesized the nano-amorphous therapy we started testing it in people with bone degeneration, where we immediately saw results. Since then, we have kept expanding our treatment focus to new areas including osteoporotic fractures, ***hypoparathyroidism, pancreatic cancer and Crohn’s disease. We have also seen promising data in osteoarthritis and skin conditions, while we believe there are opportunities to treat respiratory inflammatory conditions. As we continue to learn more about the therapy and test it in more conditions, we believe the opportunities will only continue to expand and grow.

Significant Milestones

What have been some of the company’s most significant milestones? Ben cited:

Our most significant milestones to date have been our clinical trial successes. We recently announced long-term data from our Crohn’s trial, finding that nearly two-thirds of patients achieved sustained clinical remission through 24 weeks, while all other patients demonstrated a clinically meaningful reduction in disease severity.

We have also seen compelling case reports in Stage IV pancreatic cancer and look forward to advancing these to clinical studies.

Our IND approval by the FDA was also a significant milestone, enabling us to initiate a Phase 2 clinical trial for hypoparathyroidism and putting us on track to get our first drug approval.

Customer Success Stories

Can you share any specific customer success stories? Ben highlighted:

Over the years, we have received approval to treat cancer patients through a compassionate care program as an adjunct to their existing treatment regime. One of our programs is for stage IV pancreatic cancer patients who typically have 3-6 months to live.

A 73-year-old female, with a prior history of breast cancer and diabetes, was diagnosed in December 2023 with advanced pancreatic ductal adenocarcinoma invading the stomach and vessels. She began treatment of chemotherapy combined with Amorphical’s nano-amorphous minerals. Over the following 16–19 months, PET-CT imaging demonstrated partial regression of the pancreatic lesion and complete resolution of a rectal lesion. Clinically, she reported improved well-being, weight gain and normal physical examination findings and continues maintenance chemotherapy alongside mineral support.

More than two years have passed since she was diagnosed and her condition is improving.

Funding/Revenue

Are you able to discuss funding and/or revenue metrics? Ben revealed:

We are currently positioning for our next growth phase and looking for strategic partners to help us scale. We are targeting a $90M–$110M raise to advance three indications into Phase 2 trials under FDA and EMA guidance. This capital will accelerate our clinical roadmap and bridge the gap to our planned Nasdaq IPO.

Total Addressable Market (TAM)

What total addressable market (TAM) size is the company pursuing? Ben assessed:

We are targeting different markets with each of our therapies and trials:

  • In hypoparathyroidism, we are aiming to achieve first-in-class status, opening up a market valued around $1.3B.
  • For osteoporotic fractures, we have a significant opportunity. There are around 9 million fractures annually caused by osteoporosis.
  • In Crohn’s disease, there is a $10B+ inflammatory disease market.
  • Finally, Stage IV pancreatic cancer is one of the deadliest cancers, with around 3% of patients surviving five years after diagnosis.

Differentiation From The Competition

What differentiates the company from its competition? Ben affirmed:

Most therapies for our target indications only manage symptoms. But our nano-amorphous platform modulates pH at the cellular level, addressing the underlying pathology of the disease.

Because our mechanism of action is so unique, we lack a direct competitor. Instead, we are disrupting established categories:

  • Hypoparathyroidism: We move beyond conventional supplements that involve poorly absorbed, high-dose calcium. Our technology provides superior bioavailability, maintaining calcium homeostasis more effectively.
  • Inflammatory & Oncology (Crohn’s / Pancreatic Cancer): While biologics and chemotherapies are often toxic or immunosuppressive, our therapy targets the acidic microenvironment. This supports natural immune function rather than suppressing it.

*Where current treatments offer incremental relief, we target the root cause. ***Our solution appears to be well-tolerated with a high safety profile and could potentially become a more affordable and accessible long-term option than many current and emerging treatments.

Future Company Goals

What are some of the company’s future goals? Ben concluded:

Our goals focus on advancing the clinical pipeline, expanding the reach of our technology, and scaling for commercialization.

  • Clinical Development: Following proof-of-concept, we will initiate a Phase 3 trial in Hypoparathyroidism and expand Phase 2 studies in Crohn’s and pancreatic cancer. We are projecting initial regulatory submissions by 2030. Simultaneously, we continue to broaden our pipeline through the investigation of additional indications and combination therapies targeting acidic-inflammatory conditions and oncology.
  • Operational Readiness: To support this growth, we are finalizing GMP pharma certification of our manufacturing facility this year, securing the capacity for Phase 3 trials and early commercial supply. We also plan to establish U.S. operations to streamline regulatory interactions and lead future product launches.
  • Capital & Partnerships: We are actively engaging investors to secure the capital necessary for this expansion. Simultaneously, we are advancing discussions with major biopharma partners regarding licensing and co-development opportunities.
