PDS Biotechnology has announced a strategic refocus that will prioritize development of PDS0301, its tumor-targeted immunocytokine, while discontinuing further internal investment in the Phase 3 VERSATILE-003 trial of PDS0101 and pursuing strategic partnerships or other externally funded opportunities for that program.
The decision follows a comprehensive review by PDS Biotech’s board and management of the company’s development portfolio, capital allocation priorities and long-term strategy. The company concluded that directing resources toward PDS0301 could provide a more attractive path to long-term shareholder value based on its clinical data, development opportunity and capital requirements.
PDS0301 is designed to target and remodel the tumor microenvironment, with the goal of improving the effectiveness and durability of cancer treatments. PDS Biotech believes the therapy could potentially complement a range of existing and emerging oncology treatments, including RAS pathway inhibitors, bispecific antibodies, antibody-drug conjugates and radioligand therapies.
The company’s lead development opportunity is metastatic colorectal cancer.
PDS Biotech highlighted Phase 2 clinical data generated in collaboration with the National Cancer Institute involving patients with metastatic microsatellite stable and mismatch repair-proficient colorectal cancer with liver metastases. The company reported a 71% objective response rate at six months and an 80% survival rate at 24 months.
PDS Biotech contrasted those results with reported median overall survival of less than 10 months and objective response rates below 25% for this patient population. The company also noted that more than 380 patients have been treated with PDS0301 across its clinical development program.
The company plans to advance PDS0301 into a randomized Phase 2b program designed with FDA feedback in mind.
PDS Biotech said PDS0301 has demonstrated clinical activity across multiple difficult-to-treat solid tumors as a monotherapy and in double and triple combinations. The company believes its tumor microenvironment-focused mechanism could potentially support combinations with ADCs, bispecific antibodies, radioligands, targeted therapies and immunotherapies.
Beyond metastatic colorectal cancer, PDS0301 is being evaluated in recurrent prostate cancer, metastatic castration-resistant prostate cancer, Kaposi sarcoma and HPV16-positive cancers, along with other National Cancer Institute-sponsored clinical trials.
At the same time, PDS Biotech is ending internal funding for PDS0101’s VERSATILE-003 Phase 3 trial and discontinuing the study.
The company said the decision was based on the capital, time and resources required to complete Phase 3 development and support commercialization rather than a change in its assessment of PDS0101’s underlying science or clinical data. PDS Biotech now plans to seek strategic partnerships or externally funded development opportunities for the asset.
Over the next 18 to 24 months, the company expects to advance the randomized Phase 2b PDS0301 program, evaluate strategic partnering opportunities for PDS0101 and maintain disciplined capital allocation while pursuing clinical and business milestones.
The strategic shift was accompanied by a sharp decline in PDS Biotech’s share price. The stock was trading at $0.2568, down 64.87%, in the TradingView snapshot included with the announcement.
